The retromer complex is a conserved multi-meric protein complex that operates at endosomes to regulate membrane protein sorting. The key elements of retromer are a cargo-selective complex that recognizes specific membrane proteins and a membrane-bending sorting nexin dimer that can tubulate membranes. Retromer is an evolutionarily conserved protein complex composed of the VPS26, VPS29, and VPS35 proteins that selects and packages cargo proteins into transport carriers that export cargo from the endosome. The mechanisms by which retromer is recruited to the endosome and captures cargo are unknown. We show that membrane recruitment of retromer Retromer and Parkinson's disease. Retromer has been shown to play an important role in the pathology of Parkinson's disease (PD). The different pathways shown to be affected by retromer dysfunction are illustrated and include changes in lysosomal health and protein degradation, association with accessory proteins including the WASH complex and mitochondrial health. The second, larger subunit of the retromer is known as the "cargo recognition complex" (CRC), and is made up of 3 Vps proteins in both yeast and mammalian cells: Vps26, Vps29, and Vps35 (McGough and Cullen, 2011, Seaman et al., 1998, Seaman, 2012) (Fig. 2).Vps35 shows the highest homology between yeast and mammalian (human) forms, displaying 61% similarity in nucleotide sequence, and as Retromer is a protein coat that mediates endosomal protein sorting and trafficking. It assembles on endosomes and forms tubular vesicles that return specific transmembrane proteins to the plasma membrane or to the trans -Golgi network (TGN).
